The answer to your question about the relationship between the seven deadly sins as demons and the seven deadly sins as elements of human nature is: Ooh, look! Jensen and Jared are so pretty! ;-)

Re: Heart, I think Sam was more upset about (1) the super heavy-handed parallel between his killing Madison and Dean possibly having to kill him, (2) his first point-blank "human" kill, (3) having to kill someone he had a connection with, even though I agree that they hadn't become that attached, (4) puppies, and (5) the Jess thing. Because everyone that he has sex with DIES, omg. Poor doomed, emo Sammy. *g*
But the female character thing... I don't really get it either. I've read some well reasoned complaints about Jo, but there's a whole lot of random animosity out there, too.
